What is PT-141?

(bremelanotide; melanocortin receptor agonist)

PT-141 (bremelanotide) is a melanocortin receptor agonist that increases sexual interest and arousal by activating receptors in the brain and body. It is FDA-approved as Vyleesi for women with hypoactive sexual desire disorder (HSDD). It is used off-label in men with low desire. Given as a subcutaneous injection 2–4 hours before sexual activity, PT-141 does not require daily dosing. Common side effect: nausea in the first hours (30–50% of users), often mild but sometimes requiring an antiemetic.

FDA-approved

for women with HSDD (Vyleesi); off-label use in men

PT-141 works for sexual interest; arousal and response still depend on desire and stimulation. It is not prescribed with uncontrolled high blood pressure (melanocortin agonists can raise BP). Nausea is common but usually mild.

How it works,
week by week.

  1. Day 1

A US-licensed physician reviews your goals, medical history, and medications. Your BP and cardiac history are checked.

  2. Week 1

The first dose is tested privately to confirm tolerance and response.

  3. Week 2–4

Sexual interest and arousal increase predictably on-demand.

  4. Month 2

Your physician checks in: PT-141 users discuss sexual satisfaction and any side effects.

  5. Beyond

Use as-needed; your physician reviews annually or if changes occur.

Important safety information and common side effects

PT-141 activates melanocortin receptors in the brain and body. Nausea is common (30–50%), usually mild; do not use with uncontrolled high blood pressure, recent MI, unstable angina, or severe heart disease. In men, priapism (erection lasting >4 hours) is rare but requires immediate care. Tell your physician about blood pressure medications, heart history, and any skin concerns. Stop and seek care for severe headache, chest pain, priapism, severe nausea, or signs of allergic reaction.

The research behind
PT-141.

PT-141, also called bremelanotide, acts on the desire pathway in the brain. Its one labeled use is for premenopausal women with low desire. The evidence in men is small and early.

  • Women's desire0.30 and 0.42 points

Two identical 24-week trials in 1,267 premenopausal women with low desire, using bremelanotide 1.75 mg as needed against placebo. Desire rose 0.30 points more than placebo in one trial and 0.42 in the other. The effects are small.

Kingsberg et al., Obstetrics and Gynecology 2019
  • Men, nasalsmall early study

Diamond et al. tested intranasal PT-141 in healthy men and in men with mild-to-moderate erectile dysfunction, measuring erections with RigiScan. It was a small early safety, pharmacokinetic and pharmacodynamic study, not a phase 3 trial.

Diamond et al., International Journal of Impotence Research 2004
  • Men, under the skinno trial in men

We found no published phase 3 trial of PT-141 for erectile dysfunction in men. The product sold here is 0.2 mg per dose, a lower dose than the 1.75 mg studied in women, and it has no trial of its own.

PubMed search, bremelanotide and erectile dysfunction (2026)

What the studies found

  • Desire rose 0.30 and 0.42 points more than placebo in two 24-week trials of 1,267 premenopausal women with low desire (Kingsberg et al., Obstetrics and Gynecology 2019)
  • Distress about low desire fell 0.37 and 0.29 points more than placebo in the same two trials (Kingsberg et al., Obstetrics and Gynecology 2019)
  • FDA-labeled for premenopausal women with acquired, generalized hypoactive sexual desire disorder (VYLEESI prescribing information)

Trial figures are for bremelanotide 1.75 mg given under the skin in women. The PT-141 dispensed here is 0.2 mg per dose, a different dose with no trial of its own. Nausea, flushing, headache and vomiting were common in the women's trials.

What this does not say

  • The women's trial effects are modest, and they are averages across groups.
  • No human trial supports the 0.2 mg dose sold here.
  • The label warns that the drug transiently raises blood pressure; the prescriber checks cardiovascular history before any prescription.

The research
behind it.

In trials of women with HSDD, PT-141 significantly increased sexual desire, arousal, and satisfaction compared to placebo. About 40% of women showed meaningful benefit.

Off-label use in men for low desire is growing; evidence is anecdotal and from small studies. Your physician discusses realistic expectations: PT-141 does not overcome relationship problems or performance anxiety alone.

Frequently asked questions

Contact support
How do I use PT-141?

Injected under the skin, once a day. The starting prescription is 10 units (0.2 mg) each time. Your physician sets your own dose and it is printed on your label.

How much arrives in each shipment?

1 vial at the starting strength, shipped to your door from a licensed US pharmacy.

How do I use PT-141?

Inject subcutaneously 2–4 hours before anticipated sexual activity. Effects peak around 1–2 hours after injection and last several hours. One dose per 24 hours maximum. If nausea occurs, taking an anti-nausea medication (like ondansetron) 30 minutes before PT-141 helps some people. Eat a light meal beforehand; a full meal can delay effects.

How long until I see results?

PT-141: sexual effects appear within 1–2 hours of injection, peak around 2 hours, last several hours. Results require dosing each time you want the effect.

What if I have side effects?

PT-141: nausea is common (30–50%); anti-nausea medication beforehand helps. Rare: priapism in men (erection >4 hours)—seek emergency care. Stop immediately if any severe reaction occurs.

Can I use these with other medications?

Tell your physician about all medications, especially blood pressure drugs or cardiac medications. Some combinations need monitoring.

How long do I use these?

PT-141: as-needed indefinitely, or until your sexual health goals change. Annual check-ins with your physician are recommended.
